中药的有效性已经在数千年的实践中得到了验证,为挽救人类的生命做出了重大贡献,但由于其成分过于复杂,难以通过实验证明其作用机理,使中药在现代医学中的应用受到了极大限制。实际上中药具有一定的药理作用主要是因为其中含有的活性化合物,如黄花蒿具有抗疟药效是因为其中含有抗疟活性成分青蒿素,人参被称之为 “百草之王”,具有多种药理活性,也是因为其中含有多种人参皂苷成分。随着国家对中药现代化的重视日益加深,中药单体的研究已经成为研究中药抗病机理的主要途径。

与人工合成的化合物相比,中药单体往往具有更高的生物学活性和结构多样性,更易进入细胞发挥药效,拥有更高的生物学活性。此外,针对一些特殊的困难靶点,从中药单体中筛选得到其调节剂的概率更大。因此,从天然产物库中筛选得到活性先导化合物并经结构优化,获得有成药潜力的候选化合物是目前新药研发的重要途径。
