产品描述 Liproxstatin-1 is a potent and selective inhibitor of iron death (IC50=22 nM). Liproxstatin-1 protects cells from iron death induced by iron death inducers (e.g., Erastin, RSL3).
靶点活性 Ferroptosis:22 nM
体外活性 方法:小鼠成纤维细胞 L929 用 Liproxstatin-1 (0-250 nM) 处理 24 h,使用 AquaBluer 检测细胞活力。
结果:Liproxstatin-1 以剂量依赖的方式保护免受 FINs,如 BSO (10 μM)、erastin (1 μM) 和 RSL3 (0.5 μM),而不能挽救 staurosporine (0.2 μM) 或 H2O2 (200 μM) 诱导的细胞死亡。[1]
方法:少突胶质细胞 OLN93 用 Liproxstatin-1 (1 μM) 和 RSL-3 (7.89 μM) 处理 24 h,使用 micro reduced GSH assay kit 检测 GSH 水平。
结果:与 RSL-3 组相比, Liproxstatin-1 治疗增加了 GSH 水平。[2]
体内活性 方法:为检测在体内预防动物诱导型 Gpx4 破坏的潜力,将 Liproxstatin-1 (10 mg/kg) 腹腔注射给 TAM 治疗的 CreERT2;Gpx4fl/fl 小鼠,每天一次,持续两周。
结果:Liproxstatin-1 显著延长了生存期。Liproxstatin-1 组中的 TUNEL+ 细胞数量显著减少,表Liproxstatin-1 延迟了肾小管细胞的脱铁性贫血。[1]
方法:为研究铁死亡在炎症相关认知障碍中的作用,将 Liproxstatin-1 (10 mg/kg) 腹腔注射给 C57BL/6 小鼠,每天一次,五天后进行 LPS 给药。
结果:Liproxstatin-1 改善了 LPS 诱导的认知障碍小鼠模型中的记忆缺陷。Liproxstatin-1 的保护作用与铁沉积的减轻和铁死亡相关蛋白家族 TF、xCT、Fth、Gpx4 和 FtMt 的调节有关。[3]
细胞实验 Cell viability is assessed at different time points after treatment using AquaBluer according to the manufacturer’s recommendations. Alternatively, cell death is also quantified by measuring released lactate dehydrogenase activity using the cytotoxicity detection kit (LDH). (Only for Reference)
